Yes, I have in the past and buying these to do the same thing. Easy to cut with regular scissors

Is there fiberglass inside?

The contents say "silicone." They feel like silicone - very thin and flexible. I use mine under sensitive electronics to reduce the risk of static electricity. I think it would be safe to say that there is no fiberglass inside.

if I applied one or all of these to a paper towel holder rod would it keep the paper towel roll from spinning on the rod?

Haven't done it but wouldn't be surprised if it helped. Both sides of the sheets are quite non-slip. You would only need one (of the three), as they are large 12" x 16".
