Hello, The mDP-3DP Multi Monitor Splitter supports the resolution of 1920x1080 @ 60Hz for 3 monitors. So unfortunately it would not support 1440p@144hz. Accell Support.

Q: Does this support 1080p 240hz?

I use it with 120Hz triple monitors and worked fine don’t know about 250Hz

Hello, Yes, you will have to connect both USB and mDP to your Surface Book. The Mini Displayport connection is for transferring video data and the USB connection for powering the MST device.
