Will this woofer work as a replacement speaker in Klipsch AW-650?

I don't see why not. but it is not a Injection Molded Graphite diaphragm, like the OEM you are taking out. 90 watt Rms and the one you are replacing carries 85 watt. as long it don't get wet you should be ok. under shelter. (Note : check the hole pattern ) speaker will work I have one in my PC Klipsch speaker

How can I tell which sides to connect the positive/negative speaker wires?

One has a + positive and the other has a - negative, look carefully. As far as the wires, one will be solid color (positive) a d the other will have a stripe or dashes to deignate negative.

Would these work to replace in old 2 drive advent speaker

If the size (diameter) matches and the impedance (ohms) matches what you are replacing, you're good to go. What sounds good in my Bose should do OK in an Advent.
