I have thick calluses and they squeak when I slide on the low strings. Will this eliminate or at least greatly reduce the "squeak"?

First, I have to use fine sandpaper to keep the calluses on my fingers under control! The Fast Fret will reduce the noise somewhat but the use of Elixir strings will virtually eliminate the noise. I use Elixir strings on all my guitars, acoustic and electric. And... You can get good deals on these strings on Amazon!!

is this a liquid that has to be applied with a towel, or is it a paste that applies like a tube lip chap stick?

This is a liquid. It is not like a "chap stick" as others have explained. You apply the amount you like and wipe away any excess with the included cloth or any cloth.

How are you supposed to get the bottom of the strings?

Typically, you do not get the bottom of strings. Great for string change for fret board as well. Use the cloth to clean the bottom side of strings.

How often does fast frets need to be replaced? I'm assuming they get dirty and accumulate dirt and oils over time?

I use Fast Frets when I change strings. I should probably use it more often because it works well. I have several guitars and restring each of themv3 or 4 times per year. So, together I probably apply Fast Frets 15 times per year; and on applicator lasts me about 10 years.
