How long do these stay frozen in a cooler?

It depends on the size of the meal you bring, I use it to freeze my lunch size, and it keeps the lunch cold until noon

What material is filled inside, water or gel?

Sorry I can't give you a definitive answer, but it keeps my lunch cold for hours, longer than ice cubes
