Do the keys stay in the locks when unlocked?

You can detach the keys from the lock. It works like normal locks except that these are plastic locks.

Is there more then one in the box.

There are 4 big one and 4 small one in a box

Do the locks work for the big ones? My son wants to use them to play adventure games with a toy chain.

I'm not certain what amount of pressure the locks will tolerate though they should be okay for a moderate amount from a small child.
