Do you need to remove the armrest, and if so, what are the tools and instructions to do so?

Yes you have to remove the armrest from the seat all you need is a common socket set

will it fit a 2006 RX400h model?

It didn't fit my 2004 RX330. It was too big and a totally different color and shade. Unless that doesn't matter about the fit. My arm rest are worn on the driver side. The passenger side is fine. So it looks different from the driver side.

Will it fit a 2001 Lexus RX 300?

The manufacturers information indicates that it is only intended for model years 2003-2007, or so it appears.
