will this hold a denture cleaning case?

I am not sure how big a denture cleaning case is. The pockets are not what I would describe as "deep". The second row of pockets down from the top can be made into one long pocket as the dividers are attached by Velcro strips. Hope this helps you.

When the item is unfolded and hanging from its hook, what is the distance from the top of the hook to the bottom of the item?

I took this on my trip to Alaska couple of years ago and found it very very useful. You can hang it up if you want to, but I chose to just leave it in my suitcase, and use each of the pockets for the little things that I didn't want to get lost! I found it very convenient to use, and very efficient. If you're going anywhere or even at home, you may consider purchasing one of these!

Will the TSA allow this on a plane?

This was placed in the check in luggage, however, based on the travel guidelines of the few liquid items I will say Yes, it would be fine to place in a carryon Bag.
