Q: Will this mount work with monitors that are just over 24"? Mine are 24.8, would I need to go up in size?

The maximum distance between the mounting locations of the far right and far left monitor mounting heads is 48". As long as the total width of the center monitor, plus the half width of the left monitor, plus the half width of the right monitor does not exceed 48", the monitors will fit. Please also ensure all the monitors have a mounting pattern of VESA 100 or VESA 75 and the 3 monitors weigh less than 66 lbs total.

Q: Do I have to drill a hole in my desk or clamp it to the side of my desk?

It has a weighted mount. And you can choose to mount it with a hole in the desk. It has screws to lock it in place if a hole is drilled.

Would this support a larger monitor in the center with smaller ones on the outside?  I currently have a 27" and two 21"...

I have three HP S2331 23 Inch monitors straight across in a row 21 1/2" measured across the bottom corner to corner.                                             Arm Lengths          O---------------O-----------------O------O------------O---------------O           I-------11"-----I-------11"-------I--4"--I------11"---I------11"------I           I-----------24"to center>>---------I------<24"to center------------IEach one of my monitors is 21.5 inches straight across the bottom and I have a gap of 2 1/8" between each monitor with no wrap around which makes the outer monitors kind of useless, to much effort is required to see the outer monitors so I don't use it like that I pull the outer monitors in and close the gap between them.If you are measuring 27" and 21" straight across the bottom I think your going to have a problem it will not work.  I would not go any larger than what I have three 21 1/2" inches measured across the bottom of the monitor because you will want to pull the outer monitors in to close the gap wrapping them  around so you will be able to see effortlessly.  Your setup would make the monitors convex like in a circle with this stand.
